SerumsYersinia enterocolitica, 2 ml, DENKA SEIKEN, Japan

Yersinia enterocolitica are gram-negative short sticks belonging to the Enterobacteriaceae family.

These immune sera are used to identify the groups O3, O5, O8 and O9 Y enterocolitica according to the classification of Wauters' in the agglutination reaction on the glass. These groups are considered as having a significant pathogenicity for humans. The serum contains agglutinins specific to each group indicated on the label of the vials.

Each serum is produced by hyperimination of healthy rabbits by reference strains of Wauters' provided by the National Institute of Infectious Diseases of Japan and inactivated heating at 56 ° C for 30 minutes. After removing cross-linked agglutinins by absorption, the serum was sterilized by filtration, and 0.08% azide was added as a preservative.

PRODUCT

Yersiniaenterocolitica sera are made from rabbits and contain 0.08% sodium azide as a preservative. The next serum is provided in a volume of 2 ml bottles with a dropper and ready to use volumes.

NECESSARY USE

Determination of Y.enterocolitica serogroup.

PRINCIPLE OF DEFINITION

When this reagent is mixed with Y.enterocolitica strain with an antigen corresponding to the reagent, the antigen-antibody reaction causes agglutination formation. The reaction is taken into account macroscopically to determine each serotype.

PROCEDURES

1. Materials required but not foreseen

Object glass, glass pencil, small test tubes, pipette and micropipette, bacteriological loop, physiological solution.

2. Preparation of reagents

Whey is ready for use.

3. Sample

For serotyping, cultures of microorganisms derived from pure culture and identified as Y.enterocolitica using biochemical tests should be used. If the sample consists of several strains, the serotype may be incorrectly identified.

4. Procedures

1) Mix a certain amount of bacteria culture (3-5 patches in the size of a matchhead) in 3 ml of physiological saline and use as an antigen suspension.

2) Place a drop of serum of each group of O3, O5, O8, O9 and the physiological saline (30 μL) as a control, on the cleared slide glass divided into pieces by a pencil on the glass.

3) Place a suspension of antigen (5-10 μl) to the serum and physiological solution on the slide glass.

4) Mix the reagents by tilting the slide back and forth for 1 minute, and observe the picture of agglutination. Agglutination is macroscopically observed in the light passing through the glass, including fluorescent light. First, it should be confirmed that agglutination is not detected when mixing the antigen suspension and the saline solution. Only strong agglutination, which occurs during 1 minute when reacting with each serum, should be considered as positive. Delay or weak agglutination is considered to be a negative result.

5) If the test sample is positive with the serum of the O3 group, follow steps 2) -4) described above with the polyvalent serum of groups O1 and O2. If agglutination with polyvalent serum of groups O1 and O2 is detected, the sample does not belong to the O3 serogroup. Y. enterocolitica O3 does not agglutinate with the polyvalent serum of O1a 02 groups.

Note

Although the groups O3, O5, O8 and O9 Yersinia enterocolitica are recognized as pathogenic to humans, the evaluation of the pathogenicity of the culture under investigation should not depend solely on the O-grouping as well as its biotype (see table below). O3 and O8 groups mainly belong to biotype 3 and biotype 4, while O5 and O9 groups belong to biotype 2.

2. Specificity

In a test performed in a similar way to a sensitivity test, sera are agglutinated only with the reference strain corresponding to the serotype, whereas in reactions with inappropriate reference strains, macroscopic agglutination is not observed
