Medium No. 15 (Hooten Bay Bouillon)

Purpose of the Brook Hootinger: Enterobacteriaceae differentiation by the ability to produce indole

How It Works: A liquid nutrient medium that is an analogue of the Hoetinger broth. Enriched with tryptophan, a substrate for the formation of indole. To detect indole in the medium, after the cultivation of microorganisms, Kovacha reagent is added 0.5 ml. In the presence of an indole a red color is formed.

Composition of the Hooten Broth:

Peptone enzymatic - 15 g / l

Sodium chloride - 6,5 g / l

Yeast extract - 0.5 g / l

Tryptophan - 0.0015 g / l

How to use Hooten's Broth:

1. Dissolve 22 g of medium when heated in 1 liter of purified water.

2. Boil 2-3 minutes, until complete dissolution of the components, pour over the test tubes.

3. Autoclate for 15 minutes at 121 C.

PH level: 7.3-0.2 ° at 25 ° C
