Composition
Indicator paper systems (NIBs) for the identification of microorganisms are discs of chromatographic paper containing certain amounts of a substrate in combination with an appropriate indicator, stabilized by a film-forming coating - polyvinyl alcohol.
Set No. 5 consists of 4 tests, which allow to determine the following enzymatic properties: utilization of glucose, sucrose, the presence of urease and amylase.
NIB-disks produce in vials of 50 disks. The set is completed in a cardboard pack of 4 bottles with instructions for use.
Kit No. 5 provides 50 tests.

Indications for use
It is designed to determine the biochemical properties of the corynebacter bacteria of diphtheria, isolated during bacteriological research, and to identify them before the biowar.
Allows to determine the following biochemical properties: the utilization of glucose, sucrose, starch and the presence of urease. The disk is placed in a test tube with a suitable test medium (0.9% sodium chloride solution, phosphate buffered saline) into which a full loop of a daily agar culture of microorganisms is suspended and incubated at a temperature of +37 ° C. The reaction is accounted for by changing the color of the dissolved substrate.

- Research is conducted with a clean culture;
- immersion of discs in test tubes is made with burnt tweezers;
- for obtaining clear results it is necessary to observe the temperature regime of the thermostat (37 ± 1) ° С, the pH of the nutrient media used and the treatment of the dishes.
Determination of the utilization of glucose, sucrose, starch and urease activity.
In a test tube with 0.3 ml of a sterile solution of sodium chloride solution 0.9% pH 7.3 ± 0.1, a complete culture loop was grown, grown on the surface of nutrient agar pH 7.6 ± 0.1 containing 20% ​​serum of horse or large cattle, or corynebacagar (CBA) for 18-24 hours at a temperature of (37 ± 1) ° C. Then the NIB-disk is immersed in this suspension. As a control, NIB disks immersed in test tubes with 0.3 ml of sterile sodium chloride solution of 0.9% pH 7.3 ± 0.1 serve as a control. The tubes are incubated at a temperature of (37 ± 1) ° C.

REGISTRATION OF RESULTS
The results of the analysis are recorded visually.

Precautions for use
Compliance with the "Rules of the device, safety of industrial sanitation, anti-epidemic regimen and personal hygiene when working in the laboratories (offices, departments) of the sanitary-epidemiological institutions of the Ministry of Health" (Moscow, 1981).
Neutralization. The used NIBs are neutralized by autoclaving with saturated steam at a pressure of 1.5 kG / cm2 at a temperature of (126 ± 2) ° C for 1 hour.
