1. PURPOSE

Nutrient medium for cultivation and excretion of pertussis microbial, dry (Boron-detectilag), hereinafter - Bordetaglar is intended to isolate pertussis microbes from infected material from pertussis patients and contact persons, as well as to cultivate strains of Bordetella.

2. CHARACTERISTICS

Bordetaglar is a fine powder from dark gray to black with white impregnations, which is obtained by mixing dry components. Powder is hygroscopic, photosensitive.

3. PRINCIPLE OF ACTION

The totality of the components that make up the nutrient medium ensures the growth of pertussis causative agent.

5. ANALYTICAL AND DIAGNOSTIC CHARACTERISTICS

The nutrient medium should provide for the growth of test strains of Bordetella pertussis 39, Bordetella pertussis 143, Bordetella pertussis 688, Bordetella pertussis 796 when 0.1 ml of microbial suspension is sown from a dilution of not less than 10-6 after 70-72 hours of incubation at a temperature of (36 1) C in the form of isolated convex, smooth, round, shiny gray colonies with smooth edges up to 1.0 mm in diameter.

9. CONDUCTING ANALYSIS

Studies of samples are carried out in accordance with the "Instruction on bacteriological and serological studies in pertussis and paracottus" (M., 1983).

9.1. Preparation of nutrient medium.

The preparation in the amount indicated on the label for the preparation of a specific lot of the nutrient medium is thoroughly stirred in 1 liter of distilled water, boiled for 2 minutes until complete melting of the agar, sterilized in an autoclave at a temperature of (112 ± 1) ° C for 30 min , cooled to a temperature of 45-55 ° C, poured into sterile Petri dishes with a layer of 5-6 mm, covered with lids and poured at a temperature of 18-25 ° C. After hardening, the medium is dried at a temperature of (361) C for 50-60 minutes, following the rules of aseptic.

9.2. To impart selective properties to Bordetaglar, antibiotics should be administered according to the "Instruction on bacteriological and serological studies in whooping cough and paracottus" (M., 1983).

9.3. Ready-to-use nutrient medium, poured into Petri dishes, opaque, black. Presence of dark inclusions is allowed.

9.4. Cups with the finished medium can be used for 7-10 days, provided that it is stored at a temperature of 2-8 * C.

9.5. Sowing and incubation

The inoculated plates are incubated in a non-inverted position (lid up) at a temperature of (361) ° C. The incubation time depends on the type of sample and the research objectives. As a rule, the results are recorded after 48-72 hours.
